Profile: David Knight BSc. MSc. Phd. PGCE.

Working as a scientist, David has taken responsibility for the assay development, cloning and protein engineering of novel antibody projects that focused on novel biological therapeutics.

Whilst working at UCB, he successfully cloned, humanised and expressed a wide variety of therapeutic antibodies including FDA approved Cimzia® that is currently used for the treatment of rheumatoid arthritis, FDA approved Reslizumab® that causes a significant reduction in the frequency of asthma.

He was one of two scientists employed by Domainex, a fast growing start up biotechnology company that focused on the rapid identification of novel soluble domains.

The work involved using high throughput combinatorial domain hunting technology to aid the discovery and crystallography of various therapeutic agents. In this, role he created methods that made the company commercially viable and instigated a major collaborative project that resulted in the company now having upwards of 70 employees.

David has mentored Postdoctoral scientists and completed antibody library platforn transfers to key phamceutical and biotechnology companies.

He was the first person to purify, crystallise, determine and report the initial structure of a potential neurologically important molecule (GABARAP) that was originally thought to be invloved in alzheimer's disease.

David has effectively applied a range of expression technologies to target molecules using mammalian tissue culture, yeast cell culture and microbial systems and presented data at board level whilst working at Europe's largest biotechnology company.

He has acquired a strong commercial awareness of the drug discovery business and was promoted during my time with my employer for contributions to challenging programmes.

Areas of interest include - phage display, antibody engineering, molecular biology, protein expression purification, pharmaceutical targets, science writing, science and math teaching, X-ray crystallography, web design and computer programming.
